RESUMO

Nanoparticles are known bio elicitors in plant biotechnology. Different concentrations of ZnO, CuO and CoO nanoparticles were used for the enhanced accumulation of secondary metabolites and antioxidant activities in the callus derived from root, shoot and leaf of Artemisia annua L. Biomass of callus was somehow affected on high concentrations of Nps. Phenolic content was observed maximum (60µg) in shoot callus at 0.1mg/l of CuONps. Total antioxidant activity was observed maximum (33µg) in root callus at 0.1mg/l of ZnOnps. Total reducing power maximum (33µg) was observed in root callus at concentration of 0.05 mg/l of CoONps. Maximum radical scavenging activity was observed in shoot callus at 0.05mg/l of ZnONps. Rutin gallic acid and caffic acid were also determined in most of the samples by HPLC. The study concludes that different Nps have positive effect on the induction of secondary metabolites in A.annua plant.

Diferentes concentrações de nanopartículas de ZnO, CuO e CoO foram usadas para o acúmulo aumentado de metabólitos secundários e atividades antioxidantes no calo derivado da raiz, parte aérea e folha de Artemisiaannua L. A biomassa do calo foi de alguma forma afetada em altas concentrações de Nps. O conteúdo fenólico foi observado no máximo (60 µg) no calo da parte aérea a 0,1 mg / l de CuONps. A atividade antioxidante total foi observada no máximo (33µg) no calo radicular a 0,1mg / l de ZnOnps. O poder de redução total máximo (33µg) foi observado no calo radicular na concentração de 0,05 mg / l de CoONps. Atividade máxima de eliminação de radicais foi observada no calo da parte aérea a 0,05mg / l de ZnONps. O ácido rutina-gálico e o ácido caffic também foram determinados na maioria das amostras por HPLC. O estudo conclui que diferentes Nps têm efeito positivo na indução de metabólitos secundários na planta de A.annua.

Annual ryegrass is one of the most serious, costly weeds of winter cropping systems in Australia. To determine whether its competition-mediated plant defence mechanisms effect on wheat grain quality, wheat (cv. Yitpi) and annual ryegrass were grown under two levels of CO2 (400 ppm; (a[CO2]) vs 700 ppm; (e[CO2]), two levels of water (well-watered vs drought) and two types of competition (wheat only; (W), and wheat × annual ryegrass; (W × R) with four replicates. The competition × [CO2] interaction had a significant effect on wheat grain protein content, where it was increased in W × R under both e[CO2] (+17%) and a[CO2] (+21%). Grain yield, total grain reducing power and phenolic content were significantly affected by [CO2] × drought × competition. In a summary, annual ryegrass competition significantly altered the wheat grain quality under both [CO2] levels (depending on the soil water level), while also decreasing the grain yield.

RESUMO

The in vitro antioxidant activity of pinto bean, cowpea, baby lima bean, lentil, chickpea, small red bean, red kidney bean, black kidney bean, navy bean, and mung bean extracts were investigated. Significant differences were observed in the phenolic content and the antioxidant activity amongst the legume extracts. Lentils demonstrated the highest phenolic content (47.6 mg/g), total antioxidant activity (720.68 U/g), DPPH• scavenging activity (38.5%), and total reducing power, whereas baby lima beans and navy beans had the lowest. Amongst the extracts, hydroxyl radicals (•OH) scavenging was higher in black kidney bean (85.68%) and baby lima bean (74.97%) extracts. The total antioxidant activity (r=0.84), DPPH• scavenging activity (r=0.83), and total reducing power (r=0.84) were positively correlated with the total phenolic content. However, •OH scavenging and the phenolic content were not correlated.

OBJECTIVE: To evaluate the antioxidant potential of methanolic leaf extract of Indigofera cassioides (MEIC) using various in vitro antioxidant assay systems. METHODS: Antioxidant and free radical scavenging activity of MEIC was assayed by using different in vitro models like ABTS, DPPH, nitric oxide, superoxide, hydrogen peroxide and hydroxyl radical. Reductive ability of the extract was tested by the complex formation with potassium ferricyanide. Further total phenol and flavonoid contents of the crude extract were also determined. Rutin and ascorbic acid were used as standards. RESULTS: MEIC exhibited potent and concentration dependent free radical scavenging activity in all the tested models. Reductive ability was also found to increase with increase in MEIC concentration. Total phenol and flavonoid content determination showed that the extract is rich in phenols and flavonoids. CONCLUSIONS: All the results of the in vitro antioxidant assays reveal potent antioxidant and free radical scavenging activity of the leaves of Indigofera cassioides, equivalent to that of standard ascorbic acid and rutin. This potent antioxidant activity may be attributed to its high phenolic and flavonoid contents.
